产品概述

白介素-1（IL-1）是两种蛋白的统称—IL-1α和IL-1β，是由不同基因编码的，但它们识别细胞表面相同的受体。IL-1α和IL-1β结构相似，在氨基酸水平上有25%的同源性；两种都是由分子量31kDa前体裂解成17.5kDa的蛋白质。IL-1α和IL-1β都不包含典型的疏水信号肽，但是有证据证明一些非典型路径可以分泌这些因子。大量的IL-1α以前体形式保留在胞内。一部分未修饰的IL-1α运输到细胞表面但仍与细胞相连。未修饰的膜结合IL-1α体现生物活性，邻近的细胞旁分泌IL-1。IL-1α和IL-1β与特定的受体结合后发挥其作用。

产品原理

本实验采用双抗体夹心ELISA法。抗小鼠IL-1α单抗包被于酶标板上，同时加入标本、标准品和生物素化的抗小鼠IL-1α抗体，标本、标准品中的IL-1α会与加入于孔中的生物素化的抗小鼠IL-1α抗体及包被于酶标板上的单抗结合，形成免疫复合物，游离的成分被洗去；加入辣根过氧化物酶标记的亲合素，亲合素与生物素特异性结合，游离的成分被洗去。加入显色底物（显色剂），若反应孔中有IL-1α，辣根过氧化物酶会使无色的显色剂变蓝，加终止液变黄。在450nm处测OD值，IL-1α浓度与OD₄₅₀值之间呈正相关，可通过绘制标准曲线求出标本中IL-1α浓度。

产品性能

1. 灵敏度：最小可测小鼠IL-1α达4pg/ml。

2.重复性：板内、板间变异系数均<10%。

试验所需自备仪器和试剂:

1. 酶标仪(450nm检测波长滤光片，570nm或630nm校正波长滤光片)。

2. 高精度加液器及一次性吸头：0.5-10μl，2-20μl，20-200μl，200-1000μl。

3. 37℃恒温箱、蒸馏水或去离子水、坐标纸。
