白细胞介素15(IL-15)是一种很异常的细胞因子,它和IL-2具有很多相同的生物学特性,它最先在猴肾上皮细胞系(CVI/EBNA)的培养条件下被发现,其可促进小鼠T细胞系(细胞毒性T淋巴细胞株)的有丝分裂活动。IL-15和IL-2具有相似的生物活性包括刺激自然杀伤细胞、 激活的外周血T淋巴细胞、 肿瘤浸润淋巴细胞和B细胞的生长。IL-15也是人血液中T淋巴细胞的趋化因子,也可诱导NK细胞中淋巴因子激活的杀伤活性和诱导杀伤效应细胞的产生。另外,IL-15对不参与免疫反应的细胞也有影响。 骨骼肌细胞不仅可以表达IL-15和IL-15受体mRNAs.也对IL-15也有反应。人的IL-15和猴子、小鼠的IL-15分别具有97%和73%的氨基酸序列同源性。人和猴子的IL-15都可作用于小鼠细胞。
产品原理
本实验采用双抗体夹心ELISA法。抗人IL-15单抗包被于酶标板上,同时加入标本、标准品和生物素化的抗人IL-15抗体,标本、标准品中的IL-15会与加入于孔中的生物素化的抗人IL-15 抗体及包被于酶标板上的单抗结合,形成免疫复合物,游离的成分被洗去;加入辣根过氧化物酶标记的亲合素,亲合素与生物素特异性结合,游离的成分被洗去。加入显色底物(显色剂),若反应孔中有IL-15,辣根过氧化物酶会使无色的显色剂变蓝,加终止液变黄。在450nm处测OD值,IL-15浓度与OD450值之间呈正相关,可通过绘制标准曲线求出标本中IL-15浓度。
