产品概述
白细胞介素18(IL-18)是分子量为18KD的细胞因子,最早发现在中毒性休克中,作为共刺激因子以产生干扰素γ(IFN-γ)。IL-18的生物学功能与IL-12非常相似。IL-18以没有信号肽,分子量为24KD 的前体形式被合成;有活性、成熟的IL-18必须通过切割前体产成。白细胞介素1β转换酶(ICE, caspase-1)在天冬氨酸的p1位置切开IL-18前体,产生具有生物活性的,可以从细胞内分泌成熟的IL-18。文献报道,IL-18主要来源于活化的巨噬细胞,树突状细胞,Kupffer细胞,角质形成细胞,肠上皮细胞,成骨细胞,肾上腺皮质细胞和小鼠间脑等。
产品原理
本实验采用双抗体夹心ELISA法。抗人IL-18单抗包被于酶标板上,同时加入标本、标准品和生物素化的抗人IL-18抗体,标本、标准品中的IL-18会与加入于孔中的生物素化的抗人IL-18抗体及包被于酶标板上的单抗结合,形成免疫复合物,游离的成分被洗去;加入辣根过氧化物酶标记的亲合素,亲合素与生物素特异性结合,游离的成分被洗去。加入显色底物(显色剂),若反应孔中有IL-18,辣根过氧化物酶会使无色的显色剂变蓝,加终止液变黄。在450nm处测OD值, IL-18浓度与OD450值之间呈正相关,可通过绘制标准曲线求出标本中IL-18浓度。
