产品概述
粒细胞集落刺激因子(G-CSF)是在嗜中性粒性白细胞系的造血细胞的增殖、分化和活化起重要作用的多向性细胞因子。 G-CSF是通过活化的单核细胞,巨噬细胞,成纤维细胞,内皮细胞,星型胶质细胞,成骨细胞,骨髓基质细胞等细胞产生的。除此之外,各种癌细胞系和成髓细胞白血病细胞也能组成性的产生 G-CSF。已经表明G-CSF和G-CSF受体能够通过胎盘,蜕膜和子宫内膜腺体细胞在怀孕期间产生,这表明G-CSF有可能在胎盘和蜕膜中起重要作用。
在人类中,两个G-CSF的cDNA编码207和204个氨基酸残基的前体蛋白质已经得到了分离。这两个蛋白质,除了在成熟蛋白质的N端起的35个氨基酸以后的三个氨基酸差异外, 都有一段30个氨基酸的信号肽并且有一致的氨基酸序列。
产品原理
本实验采用双抗体夹心ELISA法。抗人G-CSF单抗包被于酶标板上, 标本和标准品中的G-CSF会与单抗结合,游离的成分被洗去;加入生物素化的抗人G-CSF抗体,生物素化抗人G-CSF抗体与包被在酶标板上的抗人G-CSF单抗结合而形成免疫复合物,游离的成分被洗去;加入辣根过氧化物酶标记的亲合素,亲合素与生物素特异性结合,游离的成分被洗去。加入显色底物(显色剂),若反应孔中有G-CSF,辣根过氧化物酶会使无色的显色剂变蓝,加终止液变黄。在450nm处测OD值, G-CSF浓度与OD450值之间呈正相关,可通过绘制标准曲线求出标本中G-CSF浓度。
