TNF-α,也称为 TNFSFIA,是肿瘤坏死因子家族的成员,在炎症反应与细胞凋亡中发挥重要作用。初始合成时是26kD分子量的233个氨基酸组成II型跨膜蛋白质,包含30个氨基酸组成的胞内区, 26 个氨基酸组成的跨膜区,以及177个氨基酸组成的胞外区。TNF-α胞内通过非共价的连接形成跨膜的同源二聚体。通过 TNF-α 转换酶(TACE)的作用,从跨膜区C端释放出一个由157个氨基酸组成的可溶 TNF-α(s TNF-α 。目前,已知表达TNF-α的人类细胞有:B细胞,结肠柱状上皮细胞,NK细胞, CD3+CD56+肝自然T细胞,巨噬细胞,单核细胞,树突状细胞,CD4+ CD8+ T细胞,肥大细胞,嗜中性粒细胞,角化细胞,血浆细胞以及脂肪细胞等。
产品原理
本实验采用双抗体夹心ELISA法。抗人TNF-α单抗包被于酶标板上,标本和标准品中的TNF-α会与单抗结合形成免疫复合物,游离的成分被洗去;加入生物素化的抗人TNF-α抗体,生物素化抗人TNF-α抗体与酶标板上结合的标准品或样本中的TNF-α结合而形成免疫复合物,游离的成分被洗去;加入辣根过氧化物酶标记的亲合素,亲合素与生物素特异性结合,游离的成分被洗去。加入显色底物(显色剂),若反应孔中有TNF-α,辣根过氧化物酶会使无色的显色剂变蓝,加终止液变黄。在450nm处测OD值,TNF-α浓度与OD450值之间呈正相关,可通过绘制标准曲线求出标本中TNF-α浓度。
