产品概述
白细胞介素1(IL-1)家族分子包括IL-1α,IL-1β和IL-1受体拮抗剂。IL-1α和IL-1β结合到相同细胞表面受体因此拥有许多相同的生物学功能。除了皮肤角朊细胞,一些上皮细胞和中枢神经系统的某些细胞外,IL-1不会由健康个体的未受刺激的细胞产生。然而,当机体感受炎症感染或微生物内毒素的刺激时,巨噬细胞和其他类型细胞能够产生显著增强的IL-1表达。IL-1具有一系列生物活性,在介导免疫和炎症反应过程中起重要的作用。正常条件下IL-1的产生对于宿主受伤和感染的免疫反应是非常关键的,不恰当的延长IL-1的产生已经在多种病理条件下被发现了,包括脓毒病,类风湿性关节炎,肠炎,急性和慢性髓细胞性白血病,胰岛素依赖型糖尿病,动脉粥样硬化。
产品原理
本实验采用双抗体夹心ELISA法。抗大鼠IL-1β单抗包被于酶标板上,同时加入标本、标准品和生物素化的抗大鼠IL-1β抗体,标本、标准品中的IL-1β会与加入于孔中的生物素化的抗大鼠IL-1β抗体及包被于酶标板上的单抗结合,形成免疫复合物,游离的成分被洗去;加入辣根过氧化物酶标记的亲合素,亲合素与生物素特异性结合,游离的成分被洗去。加入显色底物(显色剂),若反应孔中有IL-1β,辣根过氧化物酶会使无色的显色剂变蓝,加终止液变黄。在450nm处测OD值, IL-1β浓度与OD450值之间呈正相关,可通过绘制标准曲线求出标本中IL-1β浓度。
