产品概述
血管内皮生长因子(Vascular endothelial growth factor)又称血管通透因子(VPF),是生长因子VEGF/PDGF家族的一个成员。VEGF基因编码8个外显子。由于不同剪切形式,产生3种同型异构体(VEGF120,VEGF164,VEGF188)。大鼠VEGF164与小鼠和人的氨基酸序列的同源性分别是98%和90%。多种细胞,包括骨骼和心肌,肝细胞,造骨细胞,嗜中性粒细胞,巨噬细胞,角朊细胞,CD34+ 细胞都能表达VEGF。在体外,VEGF是一种具有重要作用的内皮细胞有丝分裂原。在体外培养内皮细胞时,VEGF能激活磷酸脂酶C,诱导胞质内的自由Ca2+ 浓度增加。VEGF对单核细胞和造骨细胞也是化学趋化因子。在体内,VEGF能诱导血管生成和增加血管渗透性。基于这些体外与体内的活性,VEGF被认为在伤口愈合、胚胎发育、固体瘤的生长与转移等过程有重要作用。
产品原理
本实验采用双抗体夹心ELISA法。抗大鼠VEGF单抗包被于酶标板上,同时加入标本、标准品和生物素化的抗大鼠VEGF抗体,标本、标准品中的VEGF会与加入于孔中的生物素化的抗大鼠抗体及包被于酶标板上的单抗结合,形成免疫复合物,游离的成分被洗去;加入辣根过氧化物酶标记的亲合素,亲合素与生物素特异性结合,游离的成分被洗去。加入显色底物(显色剂),若反应孔中有VEGF,辣根过氧化物酶会使无色的显色剂变蓝,加终止液变黄。在450nm处测OD值, IL-1β浓度与OD450值之间呈正相关,可通过绘制标准曲线求出标本中VEGF浓度。
