产品概述
IL-1包含两种不同基因的编码的蛋白,IL-1α和IL-1β,但是这两种亚型具有25%的氨基酸序列同源性,并且它们可识别相同的细胞受体。已知的分泌IL-1的细胞有成骨细胞,单核细胞,巨噬细胞,角质形成细胞,Kupffer细胞,肝细胞,胸腺和唾液腺上皮,雪旺氏细胞,成纤维细胞和神经胶质细胞(少突神经胶质,星形胶质细胞和小胶质细胞)。IL-1α和IL-1β都是由31KDa的前体蛋白合成。在不同的物种中,成熟的IL-1α的氨基酸序列有60%到70%的保守性,已发现人的IL-1可以作用于鼠的细胞系。
产品原理
本实验采用双抗体夹心ELISA法。抗大鼠IL-1α单抗包被于酶标板上,同时加入标本、标准品和生物素化的抗大鼠IL-1α抗体,标本、标准品中的IL-1α会与加入于孔中的生物素化的抗大鼠IL-1α抗体及包被于酶标板上的单抗结合,形成免疫复合物,游离的成分被洗去;加入辣根过氧化物酶标记的亲合素,亲合素与生物素特异性结合,游离的成分被洗去。加入显色底物(显色剂),若反应孔中有IL-1α,辣根过氧化物酶会使无色的显色剂变蓝,加终止液变黄。在450nm处测OD值, IL-1α浓度与OD450值之间呈正相关,可通过绘制标准曲线求出标本中IL-1α浓度。
