产品概述
白细胞介素17(也称 CTLA-8)是T细胞表达的一种多效性细胞因子。IL-17和IL-17受体(IL-17R)结合,IL-17R不和任何已知的受体家族的成员具有同源性。IL-17的表达受活化的T细胞的限制, IL-17R 的mRNA广泛分布在组织中,在已检测过的所有细胞和组织中均检测到IL-17R的mRNA。人的IL-17R 的基因位于22号染色体上,它是由866个氨基酸组成的I型膜糖蛋白,包括293个氨基酸组成胞外结构域、21个氨基酸组成的跨膜结构域和一个525个氨基酸组成的胞质尾。人IL-17R的氨基酸序列和小鼠的IL-17R具有69%的同源性。
产品原理
本实验采用双抗体夹心ELISA法。抗人IL-17R单抗包被于酶标板上,标本和标准品中的IL-17R会与单抗结合形成免疫复合物,游离的成分被洗去;加入生物素化的抗人IL-17R抗体,生物素化抗人IL-17R抗体与酶标板上结合的标准品或样本中的IL-17R结合而形成免疫复合物,游离的成分被洗去;加入辣根过氧化物酶标记的亲合素,亲合素与生物素特异性结合,游离的成分被洗去。加入显色底物(显色剂),若反应孔中有IL-17R,辣根过氧化物酶会使无色的显色剂变蓝,加终止液变黄。在450nm处测OD值, IL-17R浓度与OD450值之间呈正相关,可通过绘制标准曲线求出标本中IL-17R浓度。
